Can a school legally prevent certain people from enrolling?

I have not committed a crime, not even for minor ones, but I have had my name and character destroyed by slander and libel online. The harmful lies were about me being a criminal and a registered sex offender. I have tried to enroll back in school but they are rejecting me and some of the students have harassed me as a result of the defamatory statements.

Do I have a case?

Change your name.

Also being known as a registered sex offender is bad. No school would want you lol. Ya man change your name

No, you don't have a case with the school unfortunately. However, you can always try and get proof that you are not a sex offender or a criminal. If you can find proof of that, there might be a chance of your record/image getting clean.

Consult with a lawyer to see about a lawsuit, then you will know if you have a strong case of not.

No case against the school, and the school can look up your criminal record so that isn't really going to matter. Court cases are PUBLIC RECORD as is the sex offender registry. Schools can not allow people if they want though, and probably feel that you would have been trouble on campus from others wanting to create it.

No business is required to do business with you, just as you are not required to do business wiht them. If you are asking about a government school, mention what state or nation interests you.
